This proposal aims at identifying new biomarkers to improve the current approach for assessing the androgen disruption activity of chemicals (type flu amide) on testicular development and function. The objectives are(I) to use a DNA micro array approach to identify among thousands of testicular genes those whose expression is altered in fatally flu amide treated rats and
(II) to delineate the potential role of the proteins encoded by these altered genes in the activities of germ cells, Sirloin and Lading cells. The project is divided in 5 work packages including that concerning the project coordination. The WP2 (DNA micro array preparation) and WP3 (tabular tissues from flu amide treated rats) are performed by the industrial partner. Once the altered genes are identified in WP2 and WP3, experiments based on the in vivo and in vitro expression of these genes and transfixion of these genes in testicular cell lines will delineate their potential role in germ cell (Spa, Sterol cell (WP5) and Lydia cell (WP6) specific activities. From these experiments, we expect to (I) identify key genes involved in testicular path physiology related to an endocrine disruption activity and (ii) provide through these relevant genes new fine biomarkers to assess the ant androgen potential of chemicals.
